X-ray crystallographic studies of lipases from the fungi Humicola lanuginosa and Rhizopus delemar challenge the two-state model of interfacial activation. The research, authored by Urszula Derewenda of the University of Alberta, reports disordered 'lid' conformations in crystals under varying conditions. These findings question the simplicity of the 'enzyme theory' for how lipases achieve maximum catalytic activity.
