This dataset supports a methodology for detecting structural changes in axonal boutons using time-lapse two-photon laser scanning microscopy. The method was validated with correlative 2PLSM and 3D electron microscopy in mouse barrel cortex, showing low false positive/negative detection rates and a high correlation (r=0.93) between 2PLSM-based bouton weights and EM-measured volumes. It includes data used to construct a probabilistic model for quantifying bouton addition, elimination, and size changes.
