A crystal structure of the enzyme VioA from Chromobacterium violaceum bound to a tryptophan analogue demonstrates the promiscuity of the violacein biosynthesis pathway. The research by Hung-En Lai from Imperial College London generated 68 new-to-nature violacein analogues by supplementing engineered E. coli with substrate analogues. This work integrates synthetic biology with synthetic chemistry to rapidly access diverse xenobiotic compounds for therapeutic discovery.
